Abstract

Mitochondria were isolated from Hartmanella which satisfied various criteria of intactness and oxidized tricarboxylic acid cycle intermediates, TMPD-ascorbate and NADH. The rates of substrate oxidation were ADP dependent; P:O ratios of 2, 1.7, and 0.7 were obtained for malate, succinate and TMPD-ascorbate respectively. Antimycin A, amytal and rotenone were tested as respiratory inhibitors; the mitochondria were rotenone-insensitive. Cytochromes (a + a3), b, and c were present and changes of their redox levels were determined spectrophotometrically in various states of respiration
